Double Beam UV/Vis Spectrophotometer, model 4260/50
The Model 4260/50 dual beam spectrophotometer is an advanced instrument with a dual beam optical system whose two detectors measure the sample and the respective reference simultaneously to optimize measurement precision.
The instrument features several working modes for quantitative analysis, kinetics, scanning, multi-component analysis and DNA/protein analysis.
With a variable bandwidth (0.5/1/2/4/5 nm), it maximizes the balance between resolution, precision and accuracy for each application and therefore meets the requirements of various techniques and regulations.
The instrument is supplied with Windows-based UV/Vis Analyst application software, which allows unlimited data storage capacity.
FUNCTIONS INTEGRATED IN THE INSTRUMENT
Start-up screen: working modes
The integrated software allows full control of the instrument without the use of a PC for photometric, quantitative, scanning, kinetic, DNA/protein analysis and multi-wavelength analysis.
Quantitative analysis (standard curves)
Up to 10 standard solutions can be used to establish the equation of the standard concentration curve.
Allows selection of up to 4 methods of standard curve fitting: linear, linear through 0, quadratic and cubic equation.
Multiple wavelength analysis
Allows up to 10 wavelength values to be entered for analysis of multiple components in a series of samples.
Wavelength sweep
The wavelength range for the sweep can be selected from 0.1, 0.2, 0.5, 1, 2 or 5 nm.
A High, Medium or Low sweep speed can be selected, varying from 100 to 3000 nm/minute.
The scanning starts from longer to shorter wavelengths so the instrument remains on standby at high wavelengths, minimising degradation of UV-sensitive samples.
Precise control of the filter and lamp changes during the scanning process means that their effects are not visible in the final result.
Further manipulation of the scanning curve includes axis re-scaling, curve tracing and peak-picking.
Kinetics
For measurements over a time interval and calculation of reaction rates.
A real-time Abs vs. Time graph is displayed on the screen.
It allows programming a waiting time up to a maximum of 12 hours and measurement time intervals of 0.5, 1, 2, 5, 10, 30 seconds and 1 min.
Subsequent manipulation of the graph allows re-scaling, curve tracing and selection of the part of the curve required for rate calculation.
The rate is calculated by using a linear algorithm before multiplying by the factor entered.
DNA/Protein analysis
DNA concentration and purity is calculated by the ratio of the absorbance at 260 nm/280 nm or 260 nm/230 nm with optional subtraction of the absorbance at 320 nm.
DNA concentration =(62.9*A260)-(36.0*A280) or (49.1*A260)-(3.48*A230)
Protein concentration = (1552*A260)-(757.3*A280) or (183*A260-75.8*A230)
